갑상선안병증 환자의 섬유모세포에 구글스테론이 미치는 치료적 영향

목적 : Inflammation, hyaluronan production and adipogenesis are main pathologic events to cause Graves’ orbitopathy (GO). Guggulsterone (GS), a phytosteroid found in the resin of the guggul plant, is well-known to have a variety of biological activities to treat inflammation such as hyperlipidemia, obesity and arthritis. We are going to find out how the GS affects the fibroblasts on GO.
방법 : Using primary cultures of orbital fibroblasts from GO patients and non-GO controls, we investigated the suppressive effect of GS on the production of interleukin(IL)-1β-induced proinflammatory cytokines and hyaluronan using quantitative real time PCR (RT-PCR), western blot and ELISA. Adipogenic differentiation was evaluated by quantification of Oil-Red O stain and by assessment of protein levels of peroxisome proliferator activator gamma (PPARγ) and CCAAT-enhancer-binding proteins (C/EBP) α and β, and sterol regulatory element-binding protein-1 (SREBP-1).
결과 : Treatment with non-cytotoxic concentration of GS resulted in a dose dependent inhibition in IL-1β induced synthesis of inflammatory cytokines, including IL-6, IL-8, MCP-1 and COX-2 at both mRNA and protein levels. Hyaluronan level was significantly suppressed by GS. GS significantly decreased formation of lipid droplets and expression of PPARγ, C/EBP α/ β and SREBP-1 dose-dependently. Pretreatment with GS attenuated the phosphorylation of IL-1β induced nuclear factor kappa B (NF-κB).
결론 : In conclusion, our data show GS has significant inhibitory effects on inflammation, production of hyaluronan and adipogenesis in orbital fibroblasts. Our study provides that the first in vitro preclinical evidence of the therapeutic effect of GS to treat GO.
