흰 쥐 망막의 뮬러 세포에서 높은 혈당수치 와 최종당화산물에 의해 유발 된 염증

목적 : The main role of Müller cells is to maintain the structural and physiological stability of retina. These cells regulate extracellular microenvironment by eliminating neurotransmitters secreted from neurons to prevent cytotoxicity and regulate potassium level through ion transport. The secretion of cytokines, chemokines and growth factors in high glucose (HG) and advanced glycation end products (AGE) – Bovine serum albumin (BSA) environments of the Müller cells, which play a key role in retina and diabetic retinopathy, was analyzed in rat primary retinal Müller cells 방법 : The Müller cells ware isolated from postnatal day 6 Sprague Dawley rat retina by mixture of Type I collagenase (60 U/ml) and trypsin (0.25%). Next day, the cells were purified by shaking culture vessel (200 rpm, 1hr). And then, the cells were incubated for 5 days after the supernatant was removed. To confirm the yield of Müller cells, these were confirmed that more than 99% of Glutamine synthetase (GS) / Glial fibrillary acidic protein (GFAP) positive cells were present by flow cytometry. 결과 : In the HG environment, the protein expressions of vimentin and GFAP were increased, whereas that of Inward-rectifier potassium channel (Kir) 4.1 and GS were decreased. The secretions of interleukin (IL) - 1β and IL-6 ware confirmed to be secreted more in the AGE-BSA treatment group than in the HG treatment group. The secretion of vascular endothelial growth factor (VEGF) was also secreted more in the AGE-BSA than in the HG. 결론 : These results indicate that primary cultured responding to various stress conditions were successfully cultured, which is the basic data of primary cultured Müller cells isolated from rat retina.
