각막 내피세포 기능 부전과 연관된 방수 단백질 생물학적 기전

목적 : To investigate biologic mechanism of aqueous humor (AH) biomarkers associated with corneal endothelial cell (CEC) function 방법 : We collected AH from 5 patients with CECD during descemet membrane endothelial keratoplasty (DMEK) and also did intraopertively from 5 patients with only cataract as an age-/sex-matched control group. CEC density was measured by using specular microscope and thickness was by ultrasound pachmeter. We used LC-MS/MS with high-pH RPLC fractionation for global proteome profiling. Proteins with expression greater than ±2-fold change in LFQ intensity were classified to differentially expressed proteins (DEPs). Bioinformatics analyses was performed with GO-BP and PPI network. 결과 : 833 proteins and 839 proteins were identified from AH in control and CECD group, respectively. 51 DEPs were up-regulated (UP-DEP) and 78 were down-regulated (DN-DEP) in AH of CECD. Those were enriched in the extracellular region, membrane-bounded vesicle, and secretory granule. UP-DEPs were mainly enriched in proteolysis, catalytic activity and immune-associated processes, while DN-DEPs in signaling and development process, cellular metabolism and nervous system-related processes. Interestingly, carbonic anhydrase 2, as known CEC pump protein, significantly decreased in CECD (p <0.001). 결론 : In the present study, we proposed AH biomarkers and their functional processes regarding corneal endothelial dysfunction. Our findings would be meaningful translational approach using an integrated proteomic analysis with liquid biopsy.
