목적 : Intraocular pressure lowering drugs is the sole glaucoma treatment currently. More direct and fundamental treatment method to protect retinal ganglion cells and optic nerve head is needed in the treatment of glaucoma. Activated astrocytes contribute to remodeling of extracellular matrix remodeling in optic nerve head (ONH). In this study, we tested effect of statin on the migration of astrocyte.
방법 : Astrocytes were cultured from Asian donor ONHs and activated by TGF- β2 after pretreatment with simvastatin, lovastatin and atorvastatin for 24 hours. After treatment statins, cell adhesion and migration were studies. The effect of statin on actin cytoskeleton organization (Phalloidin staining), cell-cell adhesions (cadherin and β-catenin western blotting) and cell migration (scratch wounding assay and MYLK western blotting) were evaluated in ONH Astrocytes.
결과 : Statin inhibit a TGF- β2-induced increase in actin cytoskeleton organization and cadherin expression. Statin also inhibit a TGF- β2-induced increase in cell migration and MYLK expression.
결론 : This study demonstrates that statins have anti-migratory effect on ONH astrocytes activated by TGF- β2 treatment.
