목적 : To investigate the serial change or fluctuation of intraocular pressure (IOP) and their role in the pathogenesis of neuronal degeneration in terms of ischemia or inflammation in diabetic retina
방법 : Adult male Sprague Dawley rats were injected with a single intraperitoneal 60 mg/kg of streptozotocin (STZ) to induce diabetes. The rats were given one drop of 1% brinzolamide ophthalmic suspension twice a day or 0.005% latanoprost ophthalmic solution once a day or saline for 8 weeks in both eyes once a day. The combined treatment group were treated with both brinzolamide and latanoprost.
결과 : Diabetic rats showed slightly increased mean IOP compared to age-matched control rats (P=0.008). Fluctuation of IOP was higher in the diabetes group than the normal control or the combined treatment group (P<0.001). Apoptosis of cells in the ganglion cell layer induced by diabetes was decreased by combined treatment (P<0.001).Increased GFAP induced by diabetes was attenuated only by combined treatment of brinzolamide and latanoprost (P<0.001). Increased expression of VEGF and ICAM-1 in diabetic retina was lowered by combined treatment (P<0.001).
결론 : After induction of diabetes, fluctuation of IOP was increased in rats. Reduction of IOP fluctuation by medical treatment relieved inflammation and neurodegeneration induced by diabetes. Improvement of unstable perfusion pressure could play a role for neuroprotection in diabetic retina.
