목적 : To investigate the effect of gene editing using clustered regularly interspaced short palindromic repeats (CRISPR)-Cas (CRISPR-associated proteins) for CTGF suppresion on conjunctival scarring in glaucoma filtering surgery in rabbits.
방법 : We performed trabeculectomy on both eyes in rabbits. At the end of surgery, we injected AAV-CRISPR while fellow eye received control solution. Animals were examed on 1st day and every weeks postoperatively, and were sacrificed at 1, 2, 3, and 4 weeks. We performed H & E stains, immunohistochemical studies including alpha-SMA, and Masson Trichrome stain to evaluate degree of fibrosis.
결과 : AAV-CRISPR treated group showed better bleb survival, less fibrosis in the subconjunctival space and sclerostomy site, less collagen deposition, less cellularity, and comparable number of goblet cells than in control eyes.
결론 : This study shows that application of CTGF gene editing material in the form of AAV-CRISPR may regulate scar tissue formation at the site of trabeculectomy by blocking TGF-β related pathways, and provide a more prolonged survival of filtering blebs and better prognosis in the clinical setting.
