인간각막에서의 Sodium Hydroxymethylglycinate(SMG)를 사용한 각막교차결합술의 효과 및 안정성

목적 : The Purpose of this Study is to evaluate the corneal endothelial cell toxicity and tissue fixation intensity using 1/4 max (9.8 mM) Sodium Hydroxymethlyglycinate(SMG) for therapeutic tissue cross-linking of the cornea. 방법 : The 4 eye was set as the control group and the other 4 eye as the treatment group. The tissue was exposed to 1/4 max (9.8 mM) SMG dissolved in a buffer solution containing 0.1 M NaHCO3 at pH 8.5 for 30 minutes. The contralateral cornea was served with solution containing 0.1 M NaHCO3 as a paired control. Live / Dead assays using calcein AM and ethidium homodimer was performed to assess endothelial cell viability using a Confocal scanning fluorescent microscope. Cross-linking efficacy was determined by thermal denaturation temperature(Tm) by differential scanning calorimetry. 결과 : Endothelial cell viability was expressed as the ratio of the live cells area /area of the live+dead cells. The mean values were 83.32±16.24 %(control) and 68.87±22.55 %(treatment) (p-value>0.05, Wilcoxon signed rank test). The thermal denaturation temperature was 61.29±2.07°C(control) and 61.84±1.61°C(treatment) (p-value = 0.05, Wilcoxon signed rank test). 결론 : Endothelial cell toxicity of SMG was not significantly different from the control group and the treatment group. The thermal denaturation temperature of the treatment group was significantly higher than that of the control group. SMG is effective cross-linking agent and relatively safe.
