Effect of Losartan on the CMV infection in human trabecular meshwork cells

목적 : Human cytomegalovirus (CMV) has been demonstrated as causes of acute, recurrent, and chronic hypertensive anterior uveitis. In hypertensive anterior uveitis, human trabecular meshwork (TM) cells are considered to be a focus of inflammation. We investigated the effects of losartan, an angiotensin II receptor antagonist, on the CMV infection in trabecular meshwork cells. 방법 : Human TM cells were infected with CMV AD169, and additionally treated with losartan, dexamethasone, ganciclovir, and TGF-β1. Viral DNA accumulation and host cell response were analyzed using real-time PCR. Levels of secreted TGF-β1 were measured by determining its concentration in conditioned medium using a commercially available sandwich enzyme-linked immunosorbent assay (ELISA) kits. 결과 : The amount of viral DNA was increased exponentially at 1-7 day post infection (dpi). Losartan treatment significantly decreased the CMV copy numbers at 7 dpi. CMV-infected TM cells showed significant increase in the concentration of TGF-β1 at 3,5, and 7 dpi. Additional treatment with dexamethasone and losartan significantly decreased the TGF-β1, whereas treatment with ganciclovir did not affect the TGF-β1 level. CMV-infected TM cells stimulated by TGF-β1 significantly increased the expression of α-SMA and CTGF, which were decreased by the additional treatment with losartan. 결론 : Losartan inhibited the expressions of TGF-β1 and fibrogenic molecules in human trabecular meshwork cells. Thus, losartan appears to have the potential to decrease trabecular meshwork fibrosis in patients with viral anterior uveitis.
