Antifibrotic effects of cherry blossom flower extract on Tenon’s fibroblasts in pterygium eyes: comparison with mitomycin C

목적 : To evaluate the antifibrotic effect of cherry blossom flower extract on primary cultured pterygeal fibroblasts and to examine the utility as a potential adjunctive treatment to reduce the pterygium recurrence rate. 방법 : Samples of human Tenon’s fibroblasts were obtained during surgeries from patients with primary pterygium. Cell toxicity, cell migration, and the expression level of α-smooth muscle actin (α-SMA) and Transforming growth factor-β (TGF-β) protein were evaluated in primary cultured pterygeal fibroblasts after treatment of cherry blossom flower extract and mitomycin C (MMC). 결과 : Cell viability after cherry blossom flower extract (0.1 µg/ml) treatment was superior to MMC (200 µg/ml) treatment (63.5±5.1% vs 50.7±3.8%; p=0.02). Both cherry blossom flower extract and MMC treatment inhibited TGF-β induced cell migration compared to control (without treatment) cells (all p<0.05). Both cherry blossom flower extract and MMC treatment significantly reduced the protein expression level of α-SMA and TGF-β (all p<0.05), and the protein level of α-SMA and TGF-β following treatment with Cherry blossom flower extract was lower than that of MMC (all p<0.05). 결론 : Our study demonstrated that cherry blossom flower extract is effective in reducing fibrosis of human pterygeal fibroblasts compared to MMC. These findings may present the basis for its use as a promising candidate drug for adjuvant therapy after surgery to prevent recurrent pterygium.
