동물에서 각막내피세포에 생체 유전자 도입: electroporation과 magnetofection의 비교

목적 : To investigate the efficiency of gene delivery into corneal endothelium using electroporation or magnetofection. 방법 : Sprague-Dawley (SD) rats were used. Green fluorescence protein (GFP)-encoded plasmid were delivered into corneal endothelium using electroporation or magnetofection. Corneal opacity was evaluated. Confocal microscopy, and histology were performed. Efficiency of in vivo transfection and inflammatory reaction was evaluated. 결과 : Corneal opacity was found in 50% of MAG-PL group and 50% of MAG group. Efficiency of GFP-expressing cells were higher in EL-PL group compared to MAG-PL group (p<0.05). Hematoxylin and eosin staining showed the healthy corneal endothelial cells without inflammatory cells in EL-PL group, EL group, PL and control group during the entire experiments period. Inflammatory cell around corneal endothelium in MAG-PL and MAG group. 결론 : Electroporation or magnetofection can deliver the gene into corneal endothelium in vivo. Electroporation is more efficient and safe method for in vivo gene delivery.
