프레드니솔론이 각막상피세포의 갱존 및 세포사에 미치는 영향 분석

목적 : To investigate the effects of prednisolone on the survival and apoptosis of corneal epithelial cells. 방법 : Human corneal epithelial cells (HCECs) were primarily cultured, and treated with various concentrations of prednisolone. The survival, proliferation, and apoptosis of the cells were measured. Also, HCECs exposed to hyperosmolar stress were treated with prednisolone, and the expression of pro-inflammatory cytokines was evaluated, 결과 : The viability of HCECs was reduced by prednisolone in a dose-dependent manner. The proliferation of HCECs was suppressed, and apoptosis was increased in response to prednisolone. The detailed investigation into the apoptosis pathway revealed that prednisolone induced the up-regulation of reactive oxygen species in HCECs that was mitigated by RU38486 (glucocorticoid receptor antagonist). Caspase 3 and 9 cleavage was increased while caspase 8 was not affected by prednisolone. Prednisolone repressed the expression of pro-inflammatory cytokines in HCECs exposed to hyperosmolar stress (1mM NaCl). 결론 : Prednisolone induces apoptosis in HCECs through an intrinsic pathway via glucocorticoid receptor. whereas it possesses the anti-inflammatory activity in HCECs. Therefore, prednisolone eyedrops should be carefully used in patients with ocular surface disorders presenting as epithelial defects and inflammation.
