자외선 유발 익상편 섬유아세포에서 재조합 인간 열쇽단백질 투여 효과

목적 : To investigate the effect of recombinant human heat shock protein 27 (rhHSP27) on the ultraviolet irradiation induced human pterygium fibroblast. 방법 : Human pterygium fibroblast were isolated and cultured from specimen of normal conjunctiva and pterygium head, which collected after surgical excision. Cultured normal conjunctival fibroblasts were exposed to 20mJ/cm2 of UVB irradiation with and without 4 ug/mL of rhHSP27 treatments. Western blot analyses of α-smooth muscle actin (α-SMA), which is a marker for myofibroblast, were performed in each experimental groups and quantified using Image Gauge 4.0 software. The values of α-SMA / GAPDH were compared. 결과 : Pterygium head fibroblast showed 2.85 ± 0.11 fold greater α-SMA level compared to normal conjunctival fibroblast (P < 0.001). When normal conjunctival fibroblast exposed to UVB irradiation without rhHSP27 treatment, the level of α-SMA was increased by 1.92 ± 0.02 times (P < 0.001). However, when normal conjunctival fibroblast exposed to UVB irradiation with rhHSP27 treatment, the fibroblast demonstrated 0.82 ± 0.04 fold smaller α-SMA level than control (P=0.036). 결론 : The rhHSP27 has inhibitory effect on UVB induced differentiation of normal conjunctival fibroblast to pterygial myofibroblast. The rhHSP27 might be used as preventive treatment option for pterygium.
