목적 : Mucin is a component of tear fluid and is essential to the wettability of the ocular surface. In this study, we aimed to evaluate the effects of 4 different dry eye medications on mucin subtype (MUC1, 4, 16, 5AC) levels in a dry eye-induced mouse model.
방법 : C57BL/6 mice were separated into 6 groups: the control, dry eye-induced group, and four groups that respectively used cyclosporin A 0.05%, rebamipide 2%, diquafusol 3%, prednisolone 1% twice a day. MUC 1, 4, 5ac, 16 and inflammatory cytokines were estimated through Q-PCR from the conjunctival cells and corneal epithelial cells. Through PAS staining, IHC, conjunctival goblet cells were analyzed. FACS staining was done for evaluating MUC1, 5ac, 16 level.
결과 : By Q-PCR, the expression of MUC1, 4 mRNA were significantly elevated in the diquafusol group(p<0.05, p<0.001). A significant increase of MUC5ac was noted in the cyclosporine group(p<0.05), while MUC 16 did not show any significant change in all groups. Conjunctival goblet cells showed the greatest recovery in the cyclosporine group(p<0.001). In the FACS analysis, MUC5ac was the highest in the cyclosporine A group(5.58%), compared to the control(1.96%). MUC1 was highest in the diquafusol group(5.21%) and MUC16 was highest in the cyclosporine A(7.01%) and diquafusol (7.02%) groups.
결론 : All of studied medications affected the degree of mucin expressions and Goblet cell density. We believe that this study gives clues that could help decide which medications would be suitable for mucin deficient types of dry eye diseases.
