동소성 눈물샘 B-세포 림프종 쥐모델에서 중간엽 줄기세포가 종양 발생에 미치는 영향

목적 : To develop and characterize a lacrimal gland B-cell lymphoma model in mice, and to investigate the effects of mesenchymal stem/stromal cells (MSCs) on tumor growth and microenvironment. 방법 : We established an orthotopic lacrimal gland lymphoma model in mice by injecting 1x10^6 syngeneic B lymphoma cells (A20 murine B lymphoma cell line) into extraorbital and intraorbital lacrimal glands. To investigate whether MSCs either suppress or promote tumor growth, we injected 1x10^5 MSCs with A20 cells simultaneously. Lacrimal glands were clinically investigated at 1, 2, 3, and 4 weeks after injection, and infiltration of tumor cells and tumor-associated immune cells were evaluated by histopathologyand flow cytometry. The levels of inflammatory cytokines were also analyzed. 결과 : The volume of extraorbital and intraorbital lacrimal glands significantly increased at 1 and 2 weeks after injection of 1x10^6 A20 cells(all p<0.01). Tumor cell infiltration was noted on histopathology and flow cytometry. MSC co-injection significantly increased lymphoma cells and tumor associated immune cells including CD4, CD11b, and CD11c cells. Also, immune regulatory cells such as Foxp3 Treg cells and myeloid derived suppressor cells were increased by MSCs. Molecular assays showed tumor-associated inflammatory cytokines, such as Il-1b and TNF-alpha, were increased by MSCs. 결론 : We established a lacrimal gland B-cell lymphoma model in mice by orthotopic injection of A20 cells. MSC co-injection created inflammatory tumor microenvironment and thereby enhanced tumor development in this model.
