실리카 나노 입자가 각막 상피 세포에 미치는 영향

목적 : To know the toxicity of SiNPs on ocular cells especially ocular surface cells. 방법 : we synthesized non-porous silica nanoparticles with 50, 100 and 150 nm sizes and evaluated the cytotoxicity induced by SiNPs (25, 50, 100 µg/ml concentration) on cultured HCECs for up to 48 hours. Cellular viability was assessed by cell counting kit-8, lactate dehydrogenase assay and terminal deoxynucleotidyl transferase assay. 결과 : SiNPs were successfully up-taken by HCECs inside cytoplasmic vacuoles as revealed by transmission electron microscopy. Cellular reactive oxygen species generation was mildly elevated, dose dependently, with SiNPs, but no significant decrease of cellular viability was observed up to concentrations of 100 µg/ml for three different sized SiNPs. Western blot assays revealed that both cellular autophagy and mammalian target of rapamycin (mTOR) pathways were activated with the addition of SiNPs. 결론 : Our findings suggested that 50, 100 and 150 nm sized SiNPs did not induce significant cytotoxicity in cultured HCECs.
