두릅 추출물의 당뇨망막증에서 NF-κB 전사인자의 O-GlcNAcylation 조절을 통한 Neuroprotection효과

목적 : The modification of proteins by O-GlcNAcylation, especially the transcription factors, is an important process for cell homeostasis. Alteration of O-GlcNAcylation participates in the pathological progression of many human diseases including diabetic complications. We hence investigated the role of OGlcNAcylation of nuclear factor-kappa B (NF-κB) in retinal ganglion cell (RGC) death, and analysed the effect of Aralia elata (AE) extract on neurodegeneration in diabetic retinopathy (DR). 방법 : C57BL/6 mice with streptozotocin (STZ)-induced diabetes were daily fed either AE extracts or control diets from the onset of diabetes. All mice were killed at 2 months after the last injection of STZ or saline, and the extent of cell death and the expression of O-GlcNAc transferase (OGT), N-acetyl D-glucosaminidase (OGA), O-GlcNAcylated proteins, and NF-κB were examined. 결과 : AE did not affect the metabolic status of the diabetic mice. In diabetic retinas, a decrease in the inner retinal thickness, together with increase in TUNEL-positive RGCs, glial activation, and active caspase-3 were blocked in the retinas of AE extract-fed mice. The expression levels of O-GlcNAcylated proteins and OGT were increased in diabetic retinas and the level of O-GlcNAcylated p65 subunit of NF-κB was higher in diabetic retinas than in control retinas. AE extract downregulated OGlcNAcylation of NF-κB and prevented neurodegeneration induced by hyperglycemia 결론 : These results indicate that O-GlcNAcylation of NF-κB is involved in neuronal degeneration and AE prevents diabetes-induced RGC apoptosis via downregulation of OGlcNAcylation of NF-κB. Hence, O-GlcNAcylation may be a new target for treatment of DR and AE may have therapeutic potential to prevent diabetes-induced retinal neurodegeneration in DR.
