목적 : To investigate the distribution of IL-7 and IL-7 receptor on the ocular surface and lacrimal glands in normal, allergic, and post-keratoplasty model mice.
방법 : The mRNA expression of mouse IL-7 (mIL-7) was analyzed in a normal mouse model (C57BL/6, 7weeks) by major organs including the cornea and lacrimal glands. Samples from each organ were gained and quantitative real-time polymerase chain reaction (qRT-PCR) was done for measuring level of mIL-7. After keratoplasty and allergic conditioning in the experimental mice (Balb/C, 9weeks), sampling was performed in the cornea, lid, and lacrimal glands for comparison and qRT-PCR was done for measurement of mIL-7.
결과 : Expression of mIL-7 is markedly elevated in organs important to the immune response, such as in the lung, spleen, intestines, and lymph nodes. Additionally, mIL-7 levels can be found not only in the cornea but also in significant quantities in the lacrimal glands. Sampling from the allergic mouse model reveals that expression of mIL-7 levels are significantly increased in the cornea, lid, lacrimal glands, and spleen compared to the normal control. Finally, post-keratoplasty cornea also has increased levels of mIL-7 expression.
결론 : IL-7 has an important role in immune response and can be found in all major organs including the cornea and lacrimal glands. This study provides verification of the importance of mIL-7 in ocular surface inflammation as the results show dramatic mIL-7 upregulation and expression in cornea, lacrimal glands, and lid in response to inflammatory conditions such as post-keratoplasty and allergic exposure.
