갑상선안병증 환자에서 miR-146a 의 역할

목적 : To investigate the role of miR-146a in the regulation of inflammation in vitro model of Graves’ orbitopathy (GO). 방법 : Expression level of miR-146a in orbital adipose tissue was compared between GO and non-GO using quantitative-PCR (Q-PCR). Effects of IL-1β on miR-146a expression were evaluated in GO orbital fibroblast by Q-PCR. Effects of miR-146a mimics and inhibitor on IL-1β induced IL-6 were analyzed by ELISA and Western blotting. 결과 : The expression of miR-146a in GO orbital fibroblast was significantly higher than that in non-GO (p<0.001). IL-1β induced a time- and concentration dependent increase in miR-146a expression, which reached a plateau at approximately 16 hr and 10 ng/mL, respectively. IL-1β induced IL-6 protein production was further increased by miR-146a inhibitors, but decreased by miR-146a mimics. 결론 : MiR-146a was up-regulated by inflammatory stress such as IL-1β and the results indicated a positive effect of miR-146a toward anti-inflammatory process. MiR-146a potentially plays a role in the regulation of inflammation in orbital fibroblast, and participates in pathogenesis of GO.
