인간 군날개 섬유모세포에서 TGF-β2의 유도로 배양된 근섬유모세포에서 사이클로스포린의 항섬유화 효과

목적 : To investigate the effects of cyclosporine A (CsA) on fibrotic markers and extracellular matrix (ECM) in TGF-β2-induced myofibroblast on primary cultured human pterygium fibroblast cells 방법 : Primary cultured human pterygium fibroblasts subjected to experiment were exposed to 100 µg/mL of CsA for 20 minutes. Cells were washed with Dulbecco phosphate-buffered saline, and then incubated with TGF-β2 5ng/ml in serum-depleted Dulbecco modified Eagle medium/F-12 medium for 24 hours. Harvested cells were fixed and stained with fibrotic and ECM marker proteins. Expression levels of fibrotic and ECM marker proteins and the corresponding mRNA transcripts were determined by western blotting and real time reverse transcription polymerase chain reaction (RT-PCR) assays, respectively. 결과 : CsA reduces a number of myofibroblasts positive cells by TGF-β2-induced. CsA also markedly inhibited TGF-β2-induced α-smooth muscle actin (α-SMA) and ECM proteins on protein and mRNA expression level. 결론 : Our results demonstrate the anti-fibrotic effect of CsA in vitro. Further studies will be necessary to elucidate the precise intracellular signal mechanism responsible for CsA-induced down-regulation of myofibroblast by TGF-β2 induced on pterygium fibroblasts.
