인간 각막상피세포에서 고삼투압으로 유도되어 발현된 TNF-α, IL-6, 그리고 HLA DR에 대한 디쿠아포솔의 항염증 효과

목적 : To evaluate the effect of diquafosol, which is a pharmaceutical drug for the treatment of dry eye disease, on anti-inflammatory response in human corneal epithelial cells 방법 : Human corneal epithelial cells (HCECs) were treated with 0.6% diquafosol, which was diluted with serum free culture media, for 30 minutes after exposed to hyperosmotic stress (500 mOsM) for 30 minutes and then incubated for 24 hours with serum free media. Harvested cells were fixed and stained with inflammatory marker proteins (TNF-α, IL-6 and HLA DR). Expression levels of TNF-α, IL-6 and HLA DR proteins were measured by Western blot and mRNA expression levels were assessed with real time (RT)-PCR. 결과 : Hyperosmotic stress-induced TNF-α, IL-6 and HLA DR positive cells were reduced by diquafosol treatment in HCECs. Increased inflammatory marker proteins and mRNAs expression by hyperosmotic stress were also decreased by diquafosol exposed condition. 결론 : Diquafosol ophthalmic solution appears to be effective in the treatment of inflammatory related ocular surface disease.
