배양된 인체 각막상피세포에서의 건조화에 의한 NLRP3 인플라마좀의 발현

목적 : To investigate whether desiccation induces the expression of NLRP3 inflammasome components in primary human corneal epithelial cells (HCECs). 방법 : HCECs were desiccated by aspirating the medium for a different duration (0 to 45 min), and the cell viability was estimated using the CCK-8 assay. Then cells were collected for RNA isolation to measure the expression of mRNA by qPCR followed by reverse transcription. Also protein levels of the NLRP3 inflammasome components were measured by immunoblotting. 결과 : Desiccation altered the cell morphology and decreased the viability of HCECs. Desiccation resulted in a dose-dependent increase in the mRNA expression of the NLRP3 gene without significant changes in the expression of ASC and caspase-1 genes. NLRP3 protein levels and the conversion of pro-caspase-1 into active caspase-1 were also significantly increased after desiccation. 결론 : Desiccation increased NLRP3 protein levels likely by inducing mRNA expression in HCECs. Also desiccation converts pro-caspase-1 to active caspase-1, suggesting that the increased IL-1β in DED patients may be from activated NLRP3 inflammasome. Our study provides an insight on the role of NLRP3 inflammasomes on the pathogenesis of dry eye disease (DED).
