목적 : We investigated the localization of aquaporin-6 (AQP6) in the rat retina, and showed that AQP6 was selectively localized in the retina ganglion cell layer (GCL) as well as the outer plexiform layer (OPL).
방법 : The optic nerve injury was performed only on the right eye of rats. The animals were sacrificed at 0, 7 and 14 days after optic nerve injury, and the eyeballs were enucleated. The eyeballs were trimmed and Serial sagittal sections, with a thickness of 10 μm, were taken from the embedded eyecups. Retinal ganglion cell and retinal Müller cell culture were performed. In Immunohistochemistry(IHC), double immunostaining for AQP6 and other cell markers, such as neuronal specific nuclear protein(NeuN), anti-glutamine synthetase(GS) was performed.The results were examined using a confocal microscope
결과 : In the control eyes, AQP6 expression was clearly detected in the OPL, also observed in the retinal GCL . Along with the gradual decrease in NeuN-positive retinal ganglion cells for two weeks after optic nerve crushing injury, anti-AQP6 antibody staining gradually decreased and finally disappeared at 14 days after treatment. Notably, AQP6 expression in the retinal ganglion cells and Müller cells was confirmed in vitro.
결론 : We showed the presence of AQP6 immunoreactivity in the retinal GCL, and it is reasonable to suppose that AQP6 may be involved in the anion regulation and participate in the electrophysiology of these cells.
