BMP4 유도인자를 이용한 인간 유도만능줄기세포로부터 윤부줄기세포로의 분화 촉진 연구

목적 : To promote the differentiation of human induced pluripotent stem (iPS) cells towards limbal progenitor cells with Bone morphogenetic protein 4 (BMP 4) treatment. 방법 : We induced limbal progenitor cells from human iPS cells with limbal specific media with treatment of BMP 4. Differentiated cells were analyzed by real time PCR and immunocytochemical assay at 4 weeks. 결과 : Human dermal fibroblast-derived iPS cells were able to give rise to the limbal progenitor cells on limbal specific culture media in vitro. Moreover, BMP 4 treatment showed significantly enhanced expression of limbal stem cell markers (ABCG2 and P63) and corneal epithelial cell markers (K12), but not increased expression of corneal epithelial cell markers (K3) at 4 weeks. 결론 : This present study demonstrate the successful reprogramming strategy for the induction of limbal progenitor cells, derived from human iPSC, using limbal specific media and BMP4 treatment. We suggests the therapeutic application of limbal progenitor cells from iPS cells for the limbal stem cell deficiency in future.
