목적 : Long-term use of topical medication is needed for glaucoma treatment. Several side effects of eye drops can be caused by preservative-containing prostaglandin analogs (PGs) agents. The purpose of this study was to evaluate the side effects of PGs with benzalkonium chloride(BAC), alternative preservatives, or no preservatives.
방법 : C57BL/6 mice were treated with topical administration of normal saline or 5μl of several PGs once a day at 9 : 00 pm for one month. Right eyes were exposed to the following drugs: BAC 0.02%, bimatoprost 0.01%(with BAC 0.02%), latanoprost 0.005%(with BAC0.02%), tafluprost 0.0015%(with/without 0.001% BAC) and travoprost 0.004% (with 0.001% Polyquad). Corneal tissues were evaluated by H&E and fluorescein staining, TNF-α, IL-6, HLA-DR, pJNK, pAKT and CD44 antibodies.
결과 : BAC or BAC-containing PGs showed significantly increased corneal fluorescein staining. In the H&E staining, epithelium of BAC or BAC-containing PGs treatment, were shown that patterns were irregular or broken. Expression levels of TNF-alpha, IL-6, HLA-DR and CD44 from BAC or BAC-containing PGs treated corneal tissues significantly increased. JNK activation also highly increased in BAC or BAC containing PGs treatment. Activation of AKT was strongly detected in PF-PG and alternatively preserved PG treated mice in contradiction to JNK activation.
결론 : This in vivo study revealed that BAC-induced cytotoxicity is significant and alternatively preserved or preservative-free glaucoma medications seem to be a reasonable and viable alternative to those preserved with BAC.
