Effect of NecroX-5, a novel necrosis inhibitor, on the survival of cultivated oral mucosal epithelial sheets.

목적 : Cell sheets are exposed to ischemic damages during the interval from procurement to attachment after transplantation. We intended to investigate the effect of NecroX-5,a novel necrosis inhibitor that shows scavenger activity against mitochondrial reactive oxygen species, on the survival of cultivated human oral mucosal epithelial sheets(CHOMES) after procurement, and it’s effect on preservation of stemness after cultivated oral mucosal epithelial sheet transplantation(COMET) 방법 : One group of CHOMES was stained with trypanblue immediately after procurement(negative control). Another was kept in culture media(CM) at room temperature for 3.5 hours after procurement(positive control). Three groups were immersed in 10, 20, and 30µm NecroX-5 for 30 minutes, and then suspended in CM for 3 hours. In vivo, 6 limbal deficient rabbits received COMET after immersion in 10 µm NecroX-5 for 30 minutes(group 1), and 6 rabbits received COMET as control(group 2). 7 days after transplantation, periodic-acid-Schiff(PAS) staining and immunohistochemistry for P63 was done. 결과 : Negative control showed a live cell percentage of 80.5%, whereas positive control showed 100% of cell death. Live to dead cell ratio was 51.5% vs 48.5% in the 10 µm NecroX-5 treated sheet. There were no goblet cells on the transplanted corneal epithelial cells in both groups. The average P63 cell count of five consecutive x400 high power fields was 44.5±12.3 in group 1 and 24.3±2.1 in group 2, with statistically significant difference between groups (p=0.010). 결론 : Our results suggest that immersion in 10µm NecroX-5 may increase the cell survival of cultivated oral mucosal cell sheets, and may increase preservation of stemness.
