목적 : Experimental autoimmune uveoretinitis (EAU) is a T helper type 1 cell-mediated autoimmune disease, which serves as a model of human chronic uveitis. Caffeic acid phenethyl ester (CAPE), an active component of honeybee propolis, has a wide range of beneficial properties. The purpose of this study was to test the therapeutic effect of CAPE in EAU mouse model.
방법 : EAU was induced in C57BL/6 mice by immunization with interphotoreceptor retinoid-binding protein (IRBP) peptide 1-20 and pertussis toxin. CAPE was administered intraperitoneal injection every day (10 mg/kg/day) from day 7 to day 16. Severity of EAU was assessed with clinical scoring using slit lamp microscope, pathology and retinal flat mount.
결과 : CAPE ameliorates the severity of EAU compared with vehicle group. Infiltration of CD4+ T cell was decreased in retina of CAPE-treated EAU mice by IHC while IRBP specific T cell proliferation was not suppressed by CAPE treatment with CFSE labeled T cell proliferation analysis.
결론 : These data suggest that CAPE is a potential therapeutic agent for EAU through its inhibition of CD4 T cell infiltration and macrophage migration.
