목적 : To investigated the wound healing effect of adiponectin eye drops following a corneal alkali burn in human corneal epithelial cells.
방법 : Simian Virus 40–immortalized human corneal epithelial cell line (HCECs) was used. Chemical burn was induced by 0.01N NaOH in HCECs. After chemical injury, HCECs was treated with 0.001% or 0.01% adiponectin, or hyaluronic acid (HA). Cell viability was measured by the MTT assay. We also observed the wound healing rate of injured HCECs with migration assay for 24 hours.
결과 : Cellular viability did not change at a concentration of 0.001% adiponectin. HCECs treated with 0.001% adiponectin showed a significant improvement in wound healing rate compared with the HA-treated groups. However, there were no significant differences between the 0.01% adiponectin and HA-treated groups. The optimal concentration of adiponectin for the proliferation of corneal epithelial cells was 0.001%.
결론 : 0.001% adiponectin reduced NaOH-induced cytotoxic effects in corneal cells significantly and increased their migratory and wound healing ability, suggesting that adiponectin treatment protects corneal epithelial cells from alkali injury.
