다양한 배지에서 인간각막내피세포의 줄기세포성에 대한 연구

목적 : To investigate stemness characteristics of cultured human corneal endothelial cells (HCECs) in various media. 방법 : HCECs were cultured in various media including media A, media B, media E, and media BE. Cell morphology was evaluated with an inverted phase-contrast microscope. Immunofluorescence staining of zonula occludens-1 (ZO-1), nestin, glial fibrillary acidic protein (GFAP), and collagen VIII alpha2 (COL8A2) was performed. Cell proliferation was assessed with a cell counting kit-8 (CCK-8) assay. 결과 : A few cultured cells stained with nestin. The cellscultured in medium A expressed GFAP. Cells in medium A assumed a fibroblast-like shape, whereas cells cultured in medium B and medium BE appeared mosaic. The fibroblast-like shape of cells in medium A changed to a mosaic pattern when they were switched to medium BE. Cell proliferation was the highest in medium A. 결론 : Cultured HCECs expressed stem cell markers, including nestin and GFAP. The expression of GFAP differed according to the culture media.
