목적 : To investigate the most appropriate medium condition for cultured human corneal endothelial stem cells (HCECs)
방법 : HCECs were cultured in media A, D, ES, and N. The morphology of cells was evaluated by inverted microscopy. Immunofluorescence staining of nestin, zo-1, COLA8A1, COL8A2, and OCT 3/4 was performed. Western blotting was performed for zo-1, COLA8A1, COL8A2, OCT3/4 and ß-catenin. Cell proliferation rate was evaluated with cell counting kit -8 (CCK-8) assay.
결과 : A few cultured cells were stained with nestin. The shape of cells cultured in media A and in medium D became fibroblast-like, whereas the shape of cells in media ES and in medium N was recovered to be mosaic pattern. Expression of ZO-1, b-catenin and nestin was higher in cells cultured in medium D, where cell proliferation rate was highest.
결론 : HCEC cultured in medium D showed more prominent expression of stem cell-associated protein and higher proliferation rate.
