쥐 망막 두께 측정에 있어서 스펙트럼영역 빛간섭단층촬영의 반복성과 신뢰도

목적 : To evaluate the repeatability and reproducibility of measuring total retinal thickness (TRT) and retinal nerve fiber layer (RNFL) thickness for the foundation of in vivo measurement of retinal thickness in SD-OCT in rat. 방법 : Retinal thickness was measured using RNFL circular scan with AutoRescan and TruTracking modes in 20 eyes of LETO rats. Three RNFL circular scan images were acquired with a brief rest between measurements for intrasession repeatability. Additional three RNFL circular scans were acquired within 1 day after first for intersession repeatability. Two experienced examiners independently measured retinal thickness above same procedure for inter-examnier reproducibility. Correlation of retinal thickness measurement of OCT and histology was measured. 결과 : For TRT, intra-session and inter-session ICCs with SD-OCT were between 0.85 and 0.93. CV ranged from 3.9 % to 4.8 %. For RNFL thickness, intra-session and inter-session ICCs with SD-OCT were between 0.61 and 0.91. Inter-examiner ICC was 0.74 for TRT and 0.70 for RNFL. The CVs were 4.4 % for TRT and 6.0% for RNFL. Pearson’s correlation coefficient was a significant positive relationship between OCT and histologically determined TRT and RNFL thickness (TRT, r2=0.61, P=0.000; RNFL, r2= 0.268, P=0.019). 결론 : Measurement of TRT and RNFL thickness show high intra-session, inter-session repeatability and inter-examiner reproducibility in in vivo using SD-OCT. This method will help to allowing longitudinal study designs, following disease processes over the time, and evaluation of therapeutic effects after experimental intervention.
