목적 : To investigate the modulation of nuclear factor kappa B (NF-kB) and tonicity response element binding protein (TonEBP) activation induced by ultraviolet radiation (UV) in cultured human lens epithelial cells( HLECs)
방법 : Human lens epithelial B-3(HLEB-3) cells exposed to 20mJ/cm2 dose of UV radiation were incubated 0, 2, 4, 8, 12, 24 hours. Cellular viability was measured by a 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Expressions of NF-kB/P65 and TonEBP were measured by western blot analysis. Translocation of NF-kB and expression of TonEBP were examined by immunohistochemistry
결과 : Cellular viability on the HLEs reduced with time after UV exposure. When HLEB-3 cells were irradiated UV, translocation of NF-kB and expression of TonEBP markedly occurred 4 hours after UV irradiation
결론 : These studies demonstrate that UV activates NF-kB in HLECs via signaling through TonEBP. The activation of TonEBP in HLECs by UV radiation may have implications for the development and progression of cataracts
