망막혈관발달에서 성상세포의 신경성장인자(NGF)에 의한 수용체 (TrkA) 활성화를 통한 VEGF 발현

목적 : Communication between the vasculature and nervous system is important during embryogenesis and development. Nerve growth factor could regulate VEGF via activation of TrkA in neurons. This study was to investigate the role of NGF and its receptor TrkA in retinal vascular development.방법 : Immunofluorescence staining was performed for TrkA, TrkB, GFAP, type IV collagen in mice of post-natal day 4, 8, 12, 16, and 26. NGF induced phosphorylation of TrkA was evaluated by Western blot. NGF induced phosphorylation of Akt was evaluated by Western blot. NGF induced VEGF transcription was evaluated by RT-PCR with/without the treatment of PI3K inhibitor in astrocytes.결과 : Differential expressions of TrkA and TrkB were found in the mouse developing retina. TrkA matched extensively with astrocyte near the vessels throughout the examined developmental period of mouse retina. NGF induced phosphorylation of TrkA at least 25ng/ml. NGF induced VEGF mRNA upregulation via PI3K/Akt pathway which was reversed by PI3K inhibitor.결론 : Our results suggest that NGF induced phosphorlyation of TrkA enhances the VEGF expression via PI3K/Akt pathway, and contributes to retinal vascular development.
